毛叶木姜子的组织培养与快速繁殖

摘  要：
为尽快建立毛叶木姜子的快繁体系,以毛叶木姜子成年优株当年生萌条的带芽茎段为外植体,进行了毛叶木姜子组培及快繁技术研究。结果表明:春季(3月)树干基部的半木质化萌条是毛叶木姜子组培的最适宜外植体;1/2MS+6-BA 1.0mg/L+NAA 0.05mg/L为启动培养的最佳培养基,组培苗萌芽率达到80%以上,芽体青绿、长势好;改良MS+6-BA 3.0mg/L+IBA 0.2mg/L为毛叶木姜子组织培养的最适增殖培养基,其有效月增殖倍数为3.8,平均芽长3.66cm;毛叶木姜子组织培养的最适给光条件为:自然光培养、光照强度3 500Lux,有效月增殖倍数可达4.8;MS+IBA 1.0mg/L+NAA 0.1mg/L+蔗糖20g/L+活性炭0.05g/L为其最佳生根培养基。其组培苗移栽至泥炭土︰黄心土(3︰1)的育苗基质中,30d后,成活率超过90%。

摘  要：
To build fast propagation system of Litsea mollis Homsl,in vitro tissue culture research was done using 1-year stem segments from adult superior individuals as explants.The results showed that half lignification sprouts sampled in March were the optimal for tissue culture.The 1/2 MS+6-BA 1.0 mg/L+NAA 0.05 mg/L was the optimal initiation culture medium,with the germination rate above 80% and vitality.The modified MS+6-BA 3.0 mg/L+IBA 0.2 mg/L could ameliorate the culture multiplication and growth of axillary bud,with the monthly proliferation rate up to 3.8 and average seedling height up to 3.66 cm.Natural light culture,3 500 Lux was the best culture illumination for multiplication,the monthly proliferation rate up to 4.8.The suitable rooting medium was MS+IBA 1.0 mg/L+NAA 0.1 mg/L+20 g/L sucrose+AC 0.05 g/L.Then these cultured seedlings were transplanted into the culture tray with peat soil︰yellow soil(3︰1),the survival rate was over 90% after 30 d.
