5-氮胞苷对水曲柳合子胚外植体状态及体胚发生的影响

摘  要:
通过在培养基中添加5-氮胞苷,研究了其对水曲柳合子胚外植体状态和体胚发生的影响。在培养开始时加入不同浓度的5-氮胞苷会抑制水曲柳愈伤组织发生和水曲柳体胚发生。各浓度处理的愈伤组织诱导率均低于未添加5-氮胞苷的对照,且差异显著。除了50.0和100.0μmol/L处理的体胚发生率与未添加5-氮胞苷的对照差异不显著外,其余的处理均与对照存在显著差异,0.5μmol/L 5-氮胞苷完全抑制了体胚的发生。在不同的培养时间段加入20.0μmol/L 5-氮胞苷对水曲柳体胚发生状态有不同的影响,第1天加入明显抑制体胚发生,第15~21天加入显著促进体胚发生。以上结果的产生可能与5-氮胞苷处理后调节了外植体的DNA甲基化水平有关。添加5-氮胞苷对外植体褐化的作用趋势与对体胚发生的影响趋势几乎相同,再一次证明了水曲柳体胚发生必然伴随着外植体褐化这一结论。这种与现有关于外植体褐化与体胚发生关系相悖的现象值得进一步深入研究。

摘  要:
An experiment was conducted to study the effect of 5-azacytidine on zygotic embryo explants and somatic embryogenesis of Fraxinus mandshurica by adding it into the culture medium.Callus induction and somatic embryogenesis of F.mandshurica were inhibited by the addition of 5-azacytidine in different concentrations at the beginning of culture.The callus induction rate of each treatment was significantly lower than that of the control without addition of 5-azacytidine.The induction rate of somatic embryos of 50.0 and 100.0μmol/L treatments was not significantly different from that of the control,but the induction rate of other treatments was significantly different from that of the control.Somatic embryogenesis of treatment in 0.5μmol/L was completely suppressed.Adding 20.0μmol/L 5-azacytidine during different periods of culture had different effects on somatic embryogenesis of F.mandshurica.Somatic embryogenesis was evidently suppressed by the addition during the first day of culture,but significantly promoted by the addition during 15 to 21 days of culture.The above results might be related to the changes of explant DNA methylation level regulated by 5-azacytidine.The influence patterns of 5-azacytidine on explant browning was almost the same as that on somatic embryogenesis,which proved our former discovery of "somatic embryogenesis of F.mandshurica is companied by explant browning".This unique pattern that is different from current results about the effect of explant browning on somatic embryogenesis is worth to be studied further.
