贴梗海棠花粉生活力与贮藏性研究

摘  要:
以贴梗海棠初开花朵为试材,研究了次甲基蓝、TTC、I2-KI、а?萘酚-联苯胺4种染色剂测定花粉生活力的效果,3种贮藏条件下(-20、4、25℃)花粉生活力的变化规律及离体培养时蔗糖、硼酸、温度对花粉萌发的影响和最佳培养基组合。结果表明:不同染色剂测定的花粉生活力差异显著,次甲基蓝(84.94%)最适合贴梗海棠花粉染色,I2-KI(44.62%)不适合该种花粉染色。贮藏温度对花粉生活力影响显著,温度越低,贮藏时间越长,各贮藏条件下,花粉活力降到50%以下的时间分别为3d(25℃)、10d(4℃)、7d(-20℃),4℃贮藏条件最佳。在一定范围内,蔗糖、硼酸和温度能促进花粉萌发,超过范围后抑制萌发,且高浓度硼酸对萌发的抑制作用比低浓度硼酸强,单因素试验下最佳蔗糖、硼酸浓度和温度分别为10%、0.03%、20℃,正交实验花粉萌发的最佳组合为8%(蔗糖)+0.03%(硼酸)+20℃(温度)。

摘  要:
Taking young bloomChaenomeles speciosa as experimental materials,four coloring agents(Methylene Blue,TTC,I2-KI and alpha-naphthol benzidine)were used to measure pollen viability.Storage characters under three different temperature conditions(-20,4and 25℃)and germination percentage under different concentrations of sucrose,boric acid and temperature were studied respectively.The results showed that pollen viability measured by different coloring agents showed significantly differences.Methylene Blue(84.94%)was the best choices for rapid determination,while the worst was I2-KI(44.62%).Storage temperatures had remarkable effects on pollen viability,that was the lower the temperature,the longer the storage time.The storage time of pollen viability percentage under 50% were 3days(25℃),10days(4℃)and 7days(-25℃),the optimum temperature for pollen storage was 4℃.Within certain limits,sucrose,boric acid and temperature could promote pollen to germinate easily.If the concentration exceeded optimum capacity,germination rate of pollen would be restrained.Moreover,high concentration of boric acid gave a severe inhibition on germination by contrast with low boric acid concentration.The optimal sucrose,boric acid concentration and temperature were 10%,0.03%,20℃respectively during single factor experiment.The optimal portfolio of solid medium verified by orthogonal test was 8%(sucrose)+0.03%(boric acid)+20℃(temperature).
