枣庄黑盖猪群体中脊椎数候选基因的多态性与遗传分析

摘  要:
为了解枣庄黑盖猪种质特性,研究利用PCR和PCR-RFLP方法对影响脊椎数的2个候选基因NR6A1和VRTN在枣庄黑盖猪群体中的多态性进行检测,并应用生物学软件进行群体遗传分析及Hardy-Weinberg平衡检验.结果表明:枣庄黑盖猪群体中NR6A1基因c.575 A>G位点有AA、AG和GG 3种基因型,AA基因型为优势基因型,VRTN基因存在ins/ins、ins/-和-/-3种基因型,缺失型基因型(-/-)为优势基因型,而且NR6A1和VRTN基因突变位点为中度多态,处于Hardy-Weinberg平衡状态.由此可见枣庄黑盖猪以前没有对NRA61和VRTN基因的突变位点进行选育,利用分子标记辅助选择可快速提高这两个基因的有利基因型频率,从而提高黑盖猪群体的脊椎数.

摘  要:
In order to get a better understanding of the germplasm characteristics of Zaozhuang heigai pigs, in this study, we performed PCR and PCR-RFLP tests to determine the polymorphism of mutation loci of genes NR6A1 and VRTN affecting vertebral number, and the genetic analyses and Hardy-Weinberg Equilibrium test.The results showed that, for the SNP locus of NR6 A1, there were three genotypes, AA, AG and GG, and AA was the dominant one. For VRTN, there were three genotypes, ins/ins, ins/-and-/-, and-/-was the dominant one.Additionally, the mutation loci of these two genes were moderate polymorphism and in Hardy-Weinberg Equilibrium state. Our study indicated that previously the mutation loci of NR6 A1 and VRTN were not selected in the population. In the future, marker assisted selection would be used to improve the frequencies of favorable genotypes of these two genes quickly.
