枸杞子中OTA污染检测方法的建立

摘  要:
建立枸杞子OTA污染的检测方法,分别用100 m L甲醇-0.1 mol/L正磷酸(10∶1)溶液、5%Na HCO3-1%PEG 8 000溶液、甲醇-水(4∶1)溶液、2%Na HCO3-15%Na Cl溶液等4种溶液提取宁夏枸杞子中OTA;取20 m L提取液过固相萃取柱(SPE),再用0.1 mol/L磷酸、双蒸水淋洗,乙酸乙酯洗脱后氮气吹干,流动相溶解;高效液相色谱法测定,条件为C18反相柱分离,乙腈-水-乙酸(99∶99∶2)为流动相,荧光检测器(激发波长330 nm,发射波长460 nm)检测。结果表明,5%Na HCO3+1%PEG 8 000溶液为最佳提取液,SPE柱纯化后,HPLC测定,平均回收率为88.6%~118.0%,变异系数为3.16%~7.54%,可以作为检测宁夏枸杞子中OTA污染的方法。

摘  要:
The method of determination OTA in wolfberry with SPE-HPLC was established in this paper. The dried wolfberry was grinded and extracted Ochratoxin A with methyl alcohol-0.1 mol / L phosphoric acid(10∶1),5% Na HCO3-1% PEG 8 000,methyl alcohol-water(4∶1) and 2% Na HCO3-15% Na Cl. After C18 solid phase extraction column(C18-SPE) has activated with5 m L Methanol,5 m L double diluted water, 5 m L Sodium hydrogen carbonate(30 g / L),the above extract 20 m L was firstly brought into C18-SPE column. Then the C18-SPE column was washed with 2 m L phosphoric acid(0.1 mol / L) and 2 m L double diluted water respectively and was eluted with 5 m L ethyl acetate subsequently. The eluent was dried with N2 gas. The residue was dissolved with 0.2 m L of mobile phase for determination. The chromatographic column was C18reversed-phase column(150mm×4.6 mm i.d.) for separation. Mobile phase was acetonitrile-water-acetic acid(99∶99∶2). A high performance liquid chromatography determined with fluorescence detection. The excitation wavelength is 330 nm. The emission wavelength is 460 nm.The recoveries of this method were between 88.6% and 118.0%,and coefficient of variation varied from 3.16% to 7.54%. This method can be used to detection OTA in wolfberry.
