野核桃枝枯病菌拮抗细菌的筛选及鉴定

摘 要:
为得到对野核桃(Juglans regia L.)枝枯病有效的生防菌株,本研究采用平板对峙培养法筛选出对野核桃枝枯病菌(Juglanconis juglandina)具有拮抗作用的细菌菌株,对其进行形态学观察和16S rDNA序列同源性分析;观察拮抗细菌对菌丝生长的影响,分析该拮抗菌株的抑菌机制。结果表明,菌株QB002对野核桃枝枯病菌(J.juglandina)的生长抑制率为88.6%,同时对苹果腐烂病菌(Cytospora mali,C.salicicola)、枣果黑斑病菌(Alternaria alternata)、李褐腐病菌(Monilinia fructicola)都有一定抑制作用。该菌为革兰氏阳性细菌,在NA培养基上单菌落呈淡黄色,形状不规则,中间有褶皱,不透明,表面不光滑。结合形态特征及16S rDNA序列,明确菌株QB002为枯草芽孢杆菌(Bacillus subtilis),该菌能使供试病原菌菌丝畸形,从而抑制其生长。

摘 要:
In order to obtain a biocontrol strain that was effective against wild walnut branch rot,this study used flat confrontation culture method to screen bacterial strains that antagonize Juglanconis juglandina.After that,morphology observation and homology analysis of 16 S rDNA sequence were performed;The effects of antagonistic bacteria on mycelial growth were observed,and the antibacterial mechanism of the antagonistic strain was analyzed.The results showed that the growth inhibition rate of strain QB002 against J. juglandina was 88.6%,and meanwhile,it had a certain inhibitory effect on Cytospora mali,C. salicicola,Alternaria alternata,and Monilinia fructicola.The bacterium was a Gram-positive bacterium,and the single colony on the NA medium was light yellow,irregular in shape,wrinkled in the middle,opaque,and the surface was not smooth.According to morphological characteristics and 16 S rDNA sequence,it was confirmed that the strain QB002 was Bacillus subtilis,which could make the mycelium of the pathogen deformed,thus inhibiting its growth.
