柔嫩艾美耳球虫免疫期间鸡盲肠扁桃体γ干扰素表达动态的检测

摘  要:
根据GenBank鸡β-actin、IFN-γ的基因序列设计引物,应用逆转录-聚合酶链反应(RT-PCR)技术克隆获得β-actin和IFN-γ基因,采用β-actin为内参的半定量方法检测IFN-γ mRNA在鸡柔嫩艾美耳球虫免疫前后不同时间的表达情况,以探讨IFN-γ的表达动态与柔嫩艾美耳球虫免疫的关系。结果显示,IFN-γ在两次免疫期间的表达量整体上呈现双峰模式,首免后第7天达到一个高峰,二免后第7天达到另一个高峰。

摘  要:
According to chicken β-actin and IFN-γ gene sequences in GenBank,primers were designed, the gene sequences were amplificated by reverse transcription-polymerase chain reaction (RT-PCR) from cecal tonsil of chicken experimentally immunized by Eimeria tenella.The expression level of IFN-γ mRNA in different time after immunization was detected by semi-quantitative RT-PCR with β-actin gene as intra-reference to study the relatioship between E.tenella immunization and the kinetic expression of IFN-γ gene in chicken cecal tonsil.The result showed that the level of expression of IFN-γ gene reached 1st peak on day 7 post-1st-immunization,and reached 2nd peak on day 7 post-2nd-immunization.
