HPLC-OPA柱后衍生离子交换法对乳粉中掺水解动物蛋白检测方法的研究

摘  要:
利用高效液相离子交换色谱法寻找乳粉中掺水解动物蛋白的特征氨基酸,对乳粉和水解动物蛋白中的17种氨基酸进行了分离测定,氨基酸浓度在50~1000μmol/L时,其峰面积和氨基酸浓度的线性相关系数均大于0.9896,且乳粉和水解动物蛋白样品的重现性较好,加标回收率分别为95.7%~104.0%、95.3%~104.3%。通过对比水解动物蛋白与乳粉的氨基酸指纹图谱,找到了甘氨酸、丙氨酸等差异显著的6种特征氨基酸用于定性鉴定,并根据模拟掺假实验,建立了乳粉中掺假水解动物蛋白的半定量检测方法。

摘  要:
This paper studied the separation and analysis of 17 amino acids in milk powder and hydrolyzed animal protein by ion exchange HPLC.When the concentration of amino acid were 50~1000μmol/L,the correlation coefficients between the peak area of amino acid and the concentration of amino acid were above 0.9896.The reproducibility of milk powder and hydrolyzed animal protein was good,the ranges of recovery of amino acids were 95.7%~104.0% and 95.3%~104.3%. According to the different finger atlas of hydrolyzed amino acids,we can find six characteristic amino acids,such as glycine,alanine.And according to the imitated adulterated test,we could establish the semiquantitative detection method of adulterated hydrolyzed animal protein in milk powder.
