96份雀麦属材料遗传多样性的ISSR分析

摘  要:
利用ISSR分子标记技术研究了来自国内外的96份雀麦属材料的遗传多样性。19个ISSR引物共扩增出109条谱带,其中106条是多态性谱带,多态性位点比率为97.25%。POPGENE 1.31软件分析结果显示:96份雀麦属材料总的基因多样性指数(Nei’s)为0.2756,Shannon信息指数为0.4257,遗传多样性水平较高;基因流(Nm)为1.3273,各种群间有一定的基因交流。用SLT_NTsys2.1e软件进行UPGMA聚类分析表明,96份材料可分为6个类群,与主成分分析结果基本一致。用Arlequin3.1软件进行AMOVA分析表明,种间的分子变异比率为15.85%,种内的分子变异比率为84.15%,种内分子变异较大,种内遗传多样性高于种间的遗传多样性。

摘  要:
Inter-simple sequence repeat(ISSR) techniques were used to identify the genetic diversity of 96 brome germplasms from China and abroad.12 species were included: Bromus inermis,B.japonicus,B.biebersteinii,B.ciliatus,B.remotiflorus,B.benekenii,B.variegates,B.rubens,B.riparius,B.rigidus,B.hordeaceus,B.ircutensis.19 ISSR primers produced a total of 109 bands,of which 106 were polymorphic bands.The average percentage of polymorphic bands was 97.25%.As analyzed by the software POPEGEN1.31,the genetic diversity of 96 brome materials was high with Nei's gene diversity of 0.2756 and Shannon's information index of 0.4257.The gene flow was 0.5995,indicating that gene exchange among populations was limited.Using SLT_NTsys2.1e software,the 96 brome materials were clustered into six groups in coefficient = 0.608 by UPGMA clustering analysis,as well as six groups by principal component analysis.These two results of the analysis were consistent basically.AMOVA analysis of 12 brome species by Arlequin3.1 software indicated that the molecular variation ratio among populations is 15.85% and within populations is 84.15%.Molecular variation within populations was higher than among populations.
