山东省主栽杨树品种的AFLP分析与鉴定

摘  要：
为了解山东省主栽杨树品种间的亲缘关系,为其分类、鉴别和良种利用提供依据,利用8对引物对23个供试品种进行了AFLP分析。结果表明,8对引物共得到扩增谱带1183条,平均多态性比例为77.3%,每对引物鉴别效率为100%;参试品种间遗传差异较大,遗传距离为0.14~0.51,平均遗传距离0.31;当相似系数为0.728,供试品种被分为8大类,聚类结果与其系谱关系一致;运用特殊谱带建立了分子检索表,用于参试品种分子水平上的鉴定。

摘  要：
In order to understand the relationship of main Populus varieties in Shandong province and give basis for their classification,identification and better use,eight pair primers were used for AFLP analysis of 23 main Populus varieties.The results showed that,1183 fragments were generated totally,and average percentage of polymorphism fragments produced by each primer was 77.3%,and the identification percentage of per primer combination was 100%.The genetic differences among tested varieties were big,genetic distance among 23 materials tested was 0.14~ 0.51 and average was 0.31.Materials tested were classed into 8 categories at similarity coefficient 0.728,system cluster results based on similarity coefficient were similar with classical taxonomy.Molecular criteria of 23 varieties was put forward according to special bands.
