基于SRAP标记的羊蹄甲不同地理群体遗传多样性分析

摘  要:
应用SRAP分子标记技术,对采集自华南地区的9个不同地理群体羊蹄甲的遗传多样性和遗传分化进行了分析,从中揭示了不同生长区域的羊蹄甲群体的遗传分化水平以及基因交流程度。结果表明:从500多对引物组合中筛选出15对SRAP引物组合用于群体遗传分析,共扩增出条带172条,其中多态性条带125条,多态性百分比为70.44%;9个群体的Nei's遗传距离在0.041 1~0.248 8之间,总的Nei's基因多样性指数为0.264 8,总的Shannon's信息指数为0.391 1,群体间的遗传分化系数Gst为0.397 9,种分化(39.79%),群体间基因流Nm为0.756 6。根据聚类分析的结果,可将群内的遗传分化(60.18%)稍大于群体间的遗传9个地区的羊蹄甲分为3大类,Ⅰ类群为香港地区,Ⅱ类群包括梅州和汕头,Ⅲ类群包括广州、深圳、韶关、云浮、东莞和海南,其分布基本上都和特定地理条件相关。本研究从其分类中可为羊蹄甲种质资源的保存、分类鉴定、引种栽培及遗传改良提供参考。

摘  要:
The genetic diversity and genetic di fferentiation of Bauhinia purpurea L. collected from 9 different geographical populations in southern China were analyzed using SRAP molecular markers to reveal the genetic differentiation and gene exchange among B. purpurea populations from different growth regions. The results showed that 15 pairs of SRAP primer combinations were screened out for population genetic analysis from more than 500 pairs of primer combinations, and 172 bands were amplified, including 125 polymorphic bands, 70.44%of polymorphic bands. The Nei's genetic distance of 9 populations ranged from 0.041 1~0.248 8, the total Nei's gene diversity index was 0.264 8, the total Shannon's information index was 0.391 1, the genetic differentiation coefficient among the populations was 0.397 9, the speciation was 39.79%, and the gene flow among populations(Nm) was 0.756 6. According to the results of cluster analysis, Bauhinia purpurea(the genetic differentiation within populations(60.18%) was a little more than that among populations) from 9 regions could be divided into three categories: Group Ⅰ was Hong Kong, Group Ⅱ included Meizhou and Shantou, Group Ⅲ included Guangzhou,Shenzhen, Shaoguan, Yunfu, Dongguan and Hainan. The distribution was mainly related to specific geographical conditions. This study could provide references for the preservation, classification, introduction, cultivation and genetic improvement of germplasm resources of B. purpurea from its classification.
