摘  要:
[目的]探索偏关苜蓿在PEG模拟干旱胁迫下,脯氨酸脱氢酶(Proline Dehydrogenase,ProDH)基因表达水平及其与脯氨酸含量、脯氨酸脱氢酶活性的关系。[方法]采用20%PEG模拟干旱胁迫处理生长四周的偏关苜蓿植株,分别胁迫0h、24h、48h和72h后,测定脯氨酸含量、脯氨酸脱氢酶活性,采用qRT-PCR法检测ProDH基因的相对表达水平。[结果]随着PEG干旱胁迫时间的延长,脯氨酸的含量逐渐增加,ProDH的活性逐渐降低,ProDH基因的表达量亦表现为增加。[结论]脯氨酸脱氢酶基因表达水平与植株ProDH活性的变化不一致,表明该基因的酶活性受到了转录和翻译水平的双重调控。

摘  要:
[Objective]The aim of present study was to explore the relationship between gene expression levels,activity of ProDH,and proline contents in Medicago Sativa cv.Pianguan under PEG stress.[Methods]The alfalfa were treated with PEG(20%)for 0 h,24 h,48 hand 72 h,and the proline contents,activities and expression of ProDH mRNA of the plants were determined using qRT-PCR.[Results]The proline contents and expression of ProDH were increased constantly with the increase of treating time,while the activity of ProDH was decreased.Gene expression of ProDH was upregulated.[Conclusion]The result showed that ProDH activity was inconsistent with expression of ProDH mRNA.In conclusion,the enzymatic activity of ProDH could be dual controlled by both transcription and translation.
