解剖采集的珠母贝精子的激活条件

摘  要:
解剖成熟珠母贝 (Pinctadamargaritifera)并采集其精子 ,在不同 pH、盐度和温度条件下进行激活实验。结果显示 ,水温 2 5℃、盐度 34、pH 8.5以下时 ,珠母贝精子的激活率与活力均很低 ,pH 9.0~ 10时激活率与活力均较高 ,pH 9.5时的激活率最高 ;在水温 2 0℃ ,盐度 34时珠母贝精子只能在pH 9.5~ 10激活 ,且激活率与活力均比 2 5℃时低。在水温 2 5℃ ,pH 9.5时 ,精子在盐度 10~ 4 5能激活 ,盐度 2 5~ 35时激活率和活力很高 ;在水温 2 0℃ ,pH 9.5时 ,精子只能在盐度 2 0~ 4 0时激活。当盐度相同 ,2 0℃时的激活率与活力比 2 5℃时低。在 pH 9.5或 10的氨海水中 ,珠母贝精子在水温 5~ 4 5℃时能激活 ,在 2 5~ 30℃时激活率较高。当温度相同 ,pH 9.5时的激活率比pH 10 .0时高。解剖得到的珠母贝精子的适宜激活 pH为 9.5、适宜盐度为 2 5~ 35、适宜温度为 2 5~ 30℃。

摘  要:
The parent Pinctada margaritifera were dissected for spermatozoa collection, and under different pH, salinity and water temperature(T W), the activating experiment was conducted. The results show that at T W 25℃, pH≤8.5 and salinity 34, the activation rate of the spermatozoa is very low and they are not vigorous, and at the same T W and salinity with pH ranging from 9.5 to 10 , the activation rate is fairly high and the individuals are very vigorous. The sperm can be activated in ammonia seawater at 20℃ only while pH ranging from 9.5 to 10. At the same pH above, the activation rate and vitality of the sperm are lower at 20℃ than those at 25℃. At 25℃,the sperm can be activated by ammonia seawater with pH 9.5 and salinity 10~45, and at the same pH the activation rate and vitality are fairly high when salinity ranges from 25 to 35 . At 20℃, the sperm can be activated by ammonia seawater at pH 9.5 only with salinity 20~40, and at the same salinity, the activation rate and vitality of the sperm are lower at 20℃than those at 25℃. In ammonia seawater of pH 9.5 or 10, the sperm can be activated at 15~40℃; the activation rate is fairly high at 25~30℃. At the same T W, the activation rate is higher at pH 9.5 than that at pH 10. The optimum activating condition for the spermatozoa from dissected parent is: pH 9.5, salinity 25~35, and seawater temperature 25~30℃.
