睾丸注射Mx-NA双基因制备抗病转基因鸡的研究

摘  要:
本研究利用精原干细胞介导法体内转染重组载体pVITRO2-Mx-NA以生产抗病转基因鸡。采用睾丸注射法将脂质体包裹的线性化质粒通过随机打点注射入公鸡睾丸,采集25、50、60、70d后试验公鸡的精液,提取基因组DNA,采用常规PCR检测外源Mx-NA联合基因的整合;转染后60d采集阳性公鸡精液人工授精母鸡,采用PCR和Western blot方法检测后代血液中外源NA-Mx联合基因的表达情况。结果表明:PCR检测证实外源Mx-NA联合基因已在精子基因组中表达,精液PCR阳性率为33.33%(2/6),且能通过体外受精在后代中检测到,后代PCR检测的阳性率为31.43%(11∕35),Western blot检测F1代阳性率为28.6%(10/35)。以上结果证明外源Mx-NA基因能稳定整合到鸡的基因组中,表明睾丸注射法是一种操作简单、有效的制备抗病转基因鸡的方法。

摘  要:
In this study,the recombinant vectors pVITRO2-Mx-NA were injected into the testis through spermatogonialstem cell-mediated method to produce the antiviral transgenic chicken. The linearization recombinant vectors pVITRO2-Mx-NA entrapped by liposome were injected into the testis randomly,and the semen of the rooster was collected atdifferent periods(25 d,50 d,60 d,70 d),then the normal hens were artificially inseminated with the above semen,PCRtechnology and Western blot was conducted to detect the integration of the exogenous NA-Mx gene in the offspring′sblood and tissue. The results were as follows:PCR detection confirmed that exogenous Mx-NA gene had been expressedin the sperm genome,the exogenous Mx-NA gene were confirmed to be expressed in the sperm genome,the positive ratioof which was 33.33%(2/6) and the positive ratio of F1 offspring was 31.43%(11/35). Western blot results showedthat the positive rate of F1 generation was 28.6%(10/35). All above was proved that the exogenous NA-Mx genescould be integrated into the chicken genome,indicating the testis injection method would be a feasible,simple approach toproduce disease-resistant transgenic chicken.
