猪OAS2抗日本脑炎病毒的活性研究

摘  要：
在亚洲和一些其他地区,日本脑炎是危害重大的人畜共患病。2'-5'寡腺苷酸合成酶(2'-5'oligoadenylate synthetase,OAS)为一种干扰素诱导的抗病毒蛋白。本试验利用PCR方法获得OAS2基因ORF并将其克隆至真核表达载体pcDNA3.1(+),获得pcDNA-pOAS2;转染PK-15细胞进行pOAS2瞬时表达,通过Real-Time PCR、间接免疫荧光试验(IFA)、噬斑形成试验检测其对日本脑炎病毒(JEV)复制的影响。结果表明,猪OAS2在PK-15细胞中的瞬时表达对JEV在细胞内的复制起一定抑制作用。本试验为下一步研究OAS2抗病毒作用奠定基础。

摘  要：
In Asia and some other regions,Japanese encephalitis is a serious zoonotic disease. 2'-5'oligoadenylate synthetase(OAS) is an antiviral protein induced by interferon(IFN). To study the antiviral effect of porcine OAS2 on Japanese encephalitis virus( JEV),the porcine OAS2 gene was obtained by PCR amplification,and then the porcine OAS2 expression vector was constructed and transfected into PK-15 cells. The effects of pOAS2 on anti-JEV replication were evaluated by Real-Time PCR,IFA and plaque formation assay. The results indicated that expression of pOAS2 in PK-15 cells had significant inhibitory effect on JEV replication. The present study paved the way for further study on the pOAS2 antiviral pathway.
