渤海湾日晒盐场细菌群落结构和多样性分析

摘  要:
采用16S rRNA基因PCR-DGGE方法,比较了我国渤海湾产盐区辽宁复州湾盐场、天津汉沽盐场和山东埕口盐场中不同盐度盐田卤水中细菌多样性与群落结构。DGGE结果表明,3个盐田卤水中细菌多样性指数(Rr)和香农维纳指数(H′)与卤水盐度密切有关,但无明显区域相关性。汉沽盐场盐田卤水样本的Rr随盐度升高有逐渐降低的趋势,大连复州湾盐场与山东埕口盐场盐田卤水样本的Rr随盐度升高呈现先升高后降低的趋势。3个盐场H’差异不大,变化趋势与Rr相似。主要DGGE条带割胶回收测序结果表明,渤海湾盐田卤水中大部分为变形菌门和拟杆菌门嗜盐细菌。典范性对应性分析显示,盐度是影响盐田卤水细菌多样性的主要环境因子,盐场地理位置对卤水细菌多样性的影响不显著。

摘  要:
In this study, we investigated the structure and diversity of the bacterial community in brine from three Bohai Bay solar saltworks, namely Liaoning Fuzhouwan saltworks, Tianjin Hangu saltworks and Shandong Chengkou saltworks, by denaturing gradient gel electrophoresis(DGGE) analysis of the PCR-amplified 16 S rRNA gene fragments. The results showed that the range-weighted richness(Rr) and Shannon-Wiener index(H) of the brine were closely related with brine salinity, and no remarkable correlation was observed with geological location. Rr value of brine in Hangu saltworks decreased with increasing salinity, whilst Rr value of brine from other two saltworks increased firstly and then decreased with increasing salinity. H' value of three saltworks had the similar tendency with Rr value. Sequencing the recovered specific DGGE bands showed that Proteobacteria and Bacteroidetes were the major recognized bacterial phyla in Bohai Bay saltworks, and most of them were halophilic bacteria. Canonical correlation analysis also indicated that salinity is the main environmental factor affecting brine bacterial diversity while geographical location had less impact on it.
