虫草HSP70基因测序技术分析

摘  要:
基于冬虫夏草全基因组测序,对其HSP70基因测序技术进行分析,以5种虫草样本为试材,比较冬虫夏草HSP70基因整序列PCR测序技术与近缘种HSP70基因分段PCR测序技术两种方法适用性。结果表明,前者进程较短,效率较高,但仅适用于冬虫夏草材料;而后者通用性较好,适用于虫草多物种的分析,但工作量较大。HSP70基因在冬虫夏草种内分化小于种间的分化,冬虫夏草种及其近缘种HSP70基因序列分析结果与Sung的虫草分类系统分类相吻合。

摘  要:
Based on the entire genome sequencing of Ophiocordyceps sinensis by using samples of 5 Cordyceps species,two methods of sequencing HSP70 gene were analyzed. One was directly PCR sequencing of HSP70 gene of O. sinensis,and the other was PCR three fragments of HSP70 gene of its allied species. The results showed that the former had a shorter process with higher efficient,and only limited to O. sinensis,while the later could be applied to many species of Cordyceps,but the workload was relatively heavy. The intraspecific differentiation of HSP70 gene in O. sinensis was less than that of its allied species. The phylogenetic analysis of HSP70 gene on O. sinensis and its allied species was consistent with Sung's system of Cordyceps.
