几种药用西红花与观赏西红花的RAPD分析

摘  要:
[目的]为分析不同种源西红花的遗传多样性和亲缘关系,利用RAPD标记技术对8种西红花基因组DNA的多态性进行分析。[方法]应用22个随机引物对8个西红花品种进行RAPD分析,并根据RAPD的扩增结果,应用NTSYS-pc 2.10e软件进行聚类分析。[结果]从22个随机引物中筛选出18个多态性较高的引物,共扩增出143条DNA条带,其中108条为多态带,占总数的75.5%,平均每个随机引物扩增的DNA带数为7.9条;在遗传相似系数0.62处将8个样品分为2类,一类为药用西红花,另一类为观赏西红花。[结论]RAPD法能有效鉴别药用西红花和观赏西红花,分子聚类结果与地理因素未见确定联系。

摘  要:
[Objective] To study genetic diversity and relationship in Crocus sat1vus,inter-simple sequence repeat(RAPD) was used to analyze the genomic DNA polymorphism of 8 C.sat1vus cultivars.[Method] The 22 random primers were applied to the RAPD analysis of 8 C.sat1vus cultivars,and according to the RAPD amplification result,the software NTSYS-pc 2.10 e was used in cluster analysis.[Result] 18 higher polymorphic ones were selected from 22 random primers;143 DNA bands were amplified,and 108 of which were polymorphic(75.5 %).The average number of DNA bands amplified from each random primer is 7.9.RAPD analysis showed all samples were classified into 2 groups at 0.62.One is medicinal C.sat1vus,and the other is ornamental C.sat1vus.[Conclusion] The method RAPD can effectively identify medicinal and ornamental C.sat1vus,and molecular clustering results don't have necessary association with geographic factors.
