丝素蛋白在胶凝时的分子结构、结晶性的探讨

摘 要:
丝素溶液在一定条件下发生胶凝作用,当调节pH值至等电点附近(pH45)即会快速胶凝。由圆二色光谱可知,丝素结构从液态时的无规卷曲转化为凝胶态时的β构象,其干胶粉末经红外吸收光谱证实为β构象。经热分析和X射线衍射分析可知,丝素凝胶具有较高的结晶性。丝素的结晶部分是由Gly,Ala,Ser,Tyr等氨基酸(其中Gly、Ala占8364%)组成,由于分子间形成氢键和结晶等而使丝素溶液成为稳定的凝胶。

摘 要:
Fibroin solution gelatinizes in certain conditions.It will gelatinize quickly when pH value is regulated to about isoelectric point (pH4.5).Based on the CD spectra,it was found that the structure of fibroin was changed from random coil to β-conformation during gelation.And the structure of dried gel powder was confirmed to be β-conformation according to the IR spectra.DTA curves and X-ray diffraction showed that fibroin gel had high degree of crystallinity.The crystalline region of fibroin consists of Gly ,Ala,Ser and Tyr,etc.(among these,Gly and Ala are 83.64 mol%).Because of the formation of inter molecular hydrogen bonds and the crystallization etc.fibroin solution became steady gel.
