泥蚶三倍体的诱导研究

摘  要:
2001年5月,用6-DMAP抑制泥蚶受精卵第二极体的释放诱导三倍体的产生。实验采用三个6-DMAP浓度梯度300、450、600μmol/L,三个处理时机即受精卵第一极体排放10、30、50%和三个处理持续时间10、15、20min进行L9(34)正交实验。结果表明6-DMAP对泥蚶三倍体诱导倍化率最高可达90%左右。倍化率最高的组合为6-DMAP浓度 600μmol/L、处理时机受精卵第一极体排放50%、处理持续时间20min。另外本实验还就受精卵发育同步性对泥蚶三倍体倍化率的影响及6-DMAP对泥蚶受精卵的毒性等问题进行了探讨。

摘  要:
The Tegillarca granosa was induced to produce triploid by inhibiting the second polar body with 6 - dimethylaminopurine. The study was carried out by using an orthogonal experiment of L9(34). The 6 - DMAP concentration was set at three levels of 300,450, 600 μmol/L; the treament initial time was set at the point when 10, 30, 50 % of the first polar body was released; the lasting time of treament was 10, 15, 20 min respectively. The result showed that the highest triploid induced rate of Tegillarca granosa could reach 90 %. The optimum condition was presented as follows: 6 - DMAP concentration 600 μmol/L, initial time when 50 % of The first polar body had been released, lasting time 20 min . The effect of development synchronism of zygote on the triploid induced rate and the poisoning influence of 6 - DMAP were also discussed in this paper.
