甘草查尔酮B体外细胞毒作用研究

摘 要:
为考察甘草查尔酮B(Licochalcone B)对不同肿瘤细胞增殖的抑制作用,初步探讨其作用机制,本研究采用MTT染色法检测甘草查尔酮B对5种肿瘤细胞人乳腺癌细胞(MCF-7)、人膀胱移行细胞癌细胞(T24)、人宫颈癌细胞(HeLa)、高转移性小鼠黑色素瘤细胞(B16F10)、小鼠黑色素瘤细胞(B16F0)的增殖抑制作用;台盼蓝拒染法测定甘草查尔酮B对B16F0细胞的致死率;相差显微镜观察细胞形态;JC-1染色测定线粒体膜电势(△Ψm)。结果表明:甘草查尔酮B(5、7.5、10、12.5、15μg/mL)可浓度依赖性地抑制5种不同肿瘤细胞的增殖,在最高浓度时,抑制率分别为67.52%、67.24%、65.41%、62.81%、68.13%;随甘草查尔酮B处理浓度的增加,B16F0细胞死亡率升高,细胞皱缩,脱落细胞增多;甘草查尔酮B可明显降低B16F0细胞的线粒体膜电势,且呈现浓度依赖性。以上结果显示:甘草查尔酮B对多种肿瘤细胞体外增殖均有明显的抑制作用,这可能与其所致的线粒体损害有关。

摘 要:
To evaluate the inhibitory effect of Licochalcone B on the growth of B16F0.MTT colorimetric assay was used to examine the cell poison of Licochalcone B on MCF-7,T24,HeLa,B16F10,B16F0 cells;Trypan blue dye exclusion was used to determine the fatality rate of B16F0 cell.Phase contrast microscope was used to observe the morphological changes of B16F0 cell.JC-1 was used to determine △Ψm.Licochalcone B(5~15 μg/mL)inhibited MCF-7,T24,HeLa,B16F10,B16F0 cells proliferation in concentration-dependent manner.The fatality rate of Licochalcone B to B16F0 cell was low;cell number reduce were observed in B16F0 cell under the Licochalcone B(5~15 μg/mL)treated groups;Licochalcone B treatment reduced membrane depolarization(△Ψm)in a dose-dependent manner.Licochalcone B inhibited B16F0 cell proliferation.
