褪黑素浓度和浸种时长对干旱胁迫下紫花苜蓿种子萌发的影响

摘  要:
为了筛选出能够促进干旱胁迫下紫花苜蓿种子萌发的最适外源褪黑素(melatonin,MT)处理方法,试验采用双因素随机区组设计方法,研究不同MT浓度(0~200 μmol/L)和浸种时长(4 h、8 h和12 h)对干旱胁迫下紫花苜蓿种子发芽率、发芽势、发芽指数和活力指数的影响,采用单变量方差分析研究MT浓度和浸种时长对干旱胁迫下紫花苜蓿种子萌发的交互影响,并采用隶属函数法对各个处理组合进行综合分析,探讨促进干旱胁迫下紫花苜蓿种子萌发最适的MT浓度和浸种时长.结果表明:MT浸种会提高干旱胁迫下紫花苜蓿种子的发芽率、发芽势、发芽指数和活力指数,低浓度(25,50,75 μmol/L)MT浸种时紫花苜蓿种子的发芽率、发芽势和活力指数会随着浸种时长的延长而升高,而高浓度(100,200 μmol/L)MT浸种时紫花苜蓿种子的活力指数会随着浸种时长的延长先升高后降低;低浓度(25,50 μmol/L)MT浸种12 h后紫花苜蓿种子的发芽指数、活力指数和发芽率(除 50 μmol/L外)均显著高于其他浸种时长(P<0.05);MT浓度为50 μmol/L浸种12 h的紫花苜蓿种子萌发能力最强.说明MT浓度和浸种时长及二者的交互作用均能提高干旱胁迫下紫花苜蓿种子的萌发指数,在本试验条件下用50 μmol/L MT浸种12 h紫花苜蓿种子萌发效果最优.

摘  要:
In order to select the optimal treatment method of exogenous melatonin(MT)for promoting seed germination of Medicago sativa under drought stress,a two-factor random block design method was used to investigate the effects of different MT concentrations(0~200 μmol/L)and immersion duration(4 h,8 h and 12 h)on seed germination rate,germination potential,germination index and vigor index of the Medicago sativa seeds under drought stress.Univariate analysis of variance was used to study the interaction effects of MT concentration and immersion duration on alfalfa seed germination under drought stress.The membership function method was used to comprehensively analyze each treatment combination to explore the optimal MT concentration and immersion duration to promote alfalfa seed germination under drought stress.The results showed that MT soaking could significantly improve the germination rate,germination potential,germination index and vitality index of Medicago sativa seeds under drought stress.The the germination rate,germination potential,and vitality index of Medicago sativa seeds increased with the extension of soaking time under low MT concentrations(25,50,75 μmol/L);while under the high MT concentrations(100 and 200 μmol/L),the vitality index of alfalfa seed increased first and then decreased with the extension of immersion duration.After low MT concentrations(25,50 μmol/L)immersion duration for 12 hours,germination index,vitality index and the germination rate(except for 50 μmol/L)of alfalfa seeds was significantly higher than that in the other soaking time(P<0.05).The comprehensive analysis of membership function showed that alfalfa seed germination was the strongest when MT concentration was 50 μmol/L for 24 h.The results indicated that the germination index of alfalfa seed under drought stress could be improved by MT concentration,soaking time and their interaction,and the best effect of seed germination of Medicago sativa was achieved when 50 μmol/L MT was soaked for 12 h under the conditions of this experiment.
