摘  要:
环鸟苷二磷酸(cyclic diguanylate,c-di-GMP)是细菌内广泛存在的一类全新的第二信使分子。具有鸟苷酸环化酶(diguanylate cyclase,DGC)活性的GGDEF结构域蛋白以及具有磷酸二酯酶(phosphodiesterases,PDE)活性的EAL或HD-GYP结构域蛋白分别负责c-di-GMP的合成与降解。细菌内存在多种类型c-di-GMP的受体,负责感知细胞内c-di-GMP浓度的变化,并启动或抑制下游特定基因的表达。这些受体包括PilZ结构域蛋白,退化的GGDEF和EAL结构域蛋白,转录因子以及核糖开关(riboswitch)。c-di-GMP参与调控细菌的多种生物学功能,如生物膜形成、运动性、胞外多糖的合成、胞外酶的分泌和三型分泌系统等,从而影响病原细菌的致病性。本文对近期c-di-GMP相关研究进展进行综述,并就其应用前景进行了讨论。

摘  要:
Cyclic diguanylate( c-di-GMP) is a novel and universal second messenger in bacteria. Cellular levels of c-di-GMP are fine-tuned through two categories of proteins,one is the diguanylate cyclases( DGC) that harbor the GGDEF domains and are responsible for the synthesis of c-di-GMP; the other is phosphodiesterases( PDE) that contain EAL or HD-GYP domains and are involved in the degradation of c-di-GMP. Several types of molecules function as c-di-GMP receptors,including PilZ-domain proteins,degenerate GGDEF or EAL domain-containing proteins,transcription factors and riboswitches. These receptors sense cellular levels of the dinucleotide and regulate the expression of specific downstream genes. The c-di-GMP molecule is involved in the regulation of various biological processes in bacteria including biofilm formation,motility,the production of exopolysaccharides,the secretion of extracellular enzymes and the expression of the type III secretion system,and thus affecting bacterial virulence. In the review,we summarized progresses on the c-di-GMP-related studies and discuss prospects of c-di-GMP in regulation of bacterial virulence.
