貉α-卫星DNA的克隆及序列分析

摘  要:
根据蓝狐、银狐及犬α-卫星DNA序列设计引物RSP1和RSP2,在貉基因组中经PCR扩增获得5条分子质量大小不同的DNA片段,分别命名为RSP1~RSP5,经克隆及测序获得14个RSP序列;序列分析表明,除RSP4外,同类RSP片段的不同序列的长度差异很小且同源性大于90%,不同种类的RSP片段的长度差异很大,最大为1 815 bp,最小为495 bp,但所有序列的3′端约500 bp序列都具有高度的同源性,大于90%;经Blast检索,所克隆的RSP序列的部分区域与蓝狐、银狐及犬α-卫星DNA序列具有约75%的同源性,证明所克隆的RSP序列为貉α-卫星DNA序列,但其单体大小及亚重复的组成还需进一步研究。

摘  要:
A pair of primers,that is RSP1 and RSP2,were designed according to the alpha satellite monomers of Arctic fox,Silver fox and domestic dog.Five bands,designated as RSP1-RSP5,were achieved by PCR amplification in the genome of raccoon dog.PCR products were cloned and sequenced,and fourteen RSP sequences were obtained.The cloned sequences were variable in length,ranging from 495 to 1 815 bp,but the sequence had highly(90%) homology to each other among the same type of RSP fragment except the RSP4 sequences.Approximately the 500 bp of the 3′ side of all the RSP sequences was similar,with 90% identity except the five RSP4 sequences.Some parts of the RSP sequence had 75% homology to the satellite monomers of Arctic fox,Silver fox and domestic dog.Sequence analyses indicated that the part alpha satellite sequences were cloned successfully from raccoon dog by PCR,but the alpha satellite monomers and the subrepeat units cannot be deduced only by PCR amplification.
