反相液相色谱法测定乌桕叶中氨基酸含量

摘  要:
以邻苯二甲醛(OPA)与9-芴基羰酰氯(FMOC-CI)作衍生剂,正缬氨酸为内标,利用二极管阵列(DAD)可变波长检测程序,采用柱前衍生RP-HPLC法测定了乌桕叶中氨基酸含量。结果表明:各种氨基酸的保留时间(Rt)、相对峰面积(Ax/AI)的RSD均较满意,回收率为93.21%~114.24%;乌桕叶中含有大量的氨基酸,总得含量在1.3000mg/mL左右,含量较高的是异亮氨酸(Ile)、谷氨酸(G lu)、天冬氨酸(Asp)、亮氨酸(Leu)和精氨酸(Arg),合计含量占总含量的60%以上。18种氨基酸中含有的人体必需氨基酸有7种,其总含量为0.5984mg/mL,占总氨基酸含量的46.03%。

摘  要:
The amino acids in Chinese tallow leaves were separated and quantified by RP-HPLC after being derived with O-phthaldialdehyde(OPA) and 9-fluorenylmethyl chloroformate(FMOC),and DAD variable-wavelength as measure program.The results showed that the RSD for remaining time and A_x/A_I of amino acids was satisfied,and the recoveries ranged from 93.21% to 114.24%.Chinese tallow leaves contained plenty of amino acids which the total concentration was 1.3000mg/mL.The main amino acids in Chinese tallow leaves were Ile,Glu,Asp,Leu,and Arg,which accounted for more than 60% of total amino acids.The 7 essential amino acids in the 18 amino acids got the amount of 0.5984mg/mL,accounted for 46.03% of the total amino acids.
