卡尼鄂拉蜂越冬工蜂咽下腺转录组学分析

摘  要:
为探究卡尼鄂拉蜂(Apis mellifera carnica)越冬工蜂在越冬、春繁和淘汰3个阶段咽下腺(HGs)的调控变化规律,对蜂群3个关键阶段工蜂咽下腺进行转录组测序,差异表达基因筛选、功能富集等一系列转录组学方法分析工蜂咽下腺活性功能基因的变化,并通过qPCR对测序结果进行验证。结果表明:在3个阶段工蜂咽下腺组织中共鉴定到21 441个基因,其中737个基因在不同阶段的组织中差异表达。对差异表达基因分析发现,蜜蜂咽下腺有30个基因与糖类和能量代谢、转录因子等功能相关,其中在蜜蜂春繁阶段高表达15个,越冬阶段7个,淘汰阶段3个。这一研究提示季节性介导的咽下腺活动变化与冬春季节期间数百个基因的表达水平和模式变化显著相关。

摘  要:
In order to explore the regulation and change rules of hypopharyngeal gland(HGs) of Apis mellifera carnica during the three stages of overwintering, spring blooming and elimination, the transcriptome sequencing of worker bee HGs in three key periods of bee colony, screening of differentially expressed genes, functional enrichment, and a series of transcriptomics methods were used to analyze the changes of the functional gene of hypopharyngeal gland in worker bees, and the sequencing results were verified by qPCR. The results showed that 21 441 genes were identified in the hypopharyngeal gland tissue of worker bees in three periods, of which 737 genes were differentially expressed in different stages of tissues. Further analysis of the differentially expressed genes revealed that there were 30 genetically related differences in function between the hypopharyngeal gland in metabolism of carbohydrates and energy, transcription factors, etc. Among them, 15 were highly expressed in the spring blooming stage of the bees, seven in the overwintering stage, and three in the elimination stage. This study suggested that season-mediated changes in HG activity were significantly correlated with the expression levels and patterns of hundreds of genes during the winter-spring cycle.
