亚洲百合“欣赏”再生体系的建立

摘  要:
[目的]研究亚洲百合"欣赏"品种无菌叶片的再生体系,为"欣赏"百合种球生产国产化提供技术支持。[方法]以亚洲百合"欣赏"品种无菌芽作为材料,以MS添加不同浓度6-BA和NAA为诱导无菌叶片分化培养基,以叶片不同部位分别培养,以MS添加不同浓度6-BA为诱导愈伤组织分化的培养基,MS添加不同浓度糖和多效唑为生根结鳞茎培养基,进行组织培养。[结果]结果表明:MS+2.0 mg/L 6-BA+0.2 mg/L NAA为最佳诱导培养基。同一叶片的不同部位诱导率不同,诱导率从高到低依次是:基部>中部>顶部。2.0~3.0 mg/L6-BA对愈伤组织的分化有显著的诱导效果,分化率最高达96.4%。不同糖浓度诱导无菌芽生根结鳞茎最高达95.8%,鳞茎最大直径可达1.3 cm,根青绿色,粗壮,效果比多效唑的好。[结论]通过"欣赏"无菌叶片再生体系的建立,得到可用于生产的组培苗,该技术可应用于"欣赏"百合种苗工厂化生产。

摘  要:
[Objective] Study on Asian Lily "Xinshang" Regeneration System from breed sterile blade to it's bulb production localization to provide technical support. [Method] Used Asian Lily‘Xinshang'variety sterile buds as the material, applied MS adding different concentrations of 6-BA and NAA to induce buds,differentiation medium sterile blade to different parts of the blade were cultured, and MS adding different concentrations of 6-BA to induce more medium wound tissue differentiation, then MS adding different concentrations of sugar and MET for the rooting medium knot bulbs, for tissue culture. [Result]The results showed that: MS + 2.0mg/L 6-BA + 0.2 mg/L NAA is the best induction medium. Induction rate in different parts of the same blade of different induction rate in descending order are: base> middle> top. 2.0 ~ 3.0 mg /L6-BA on callus differentiation induced a significant effect, the highest differentiation rate reached 96.4%.Different sugar concentrations induced sterile rooting knot bulb up to 95.8%, the maximum bulb diameter up to 1.3 cm, root green, thick, the effect was better than MET. [Conclusion]By ‘Xinshang'of Leaf Regeneration System to establish a sterile, obtained for the production of tissue culture, the technology can be applied to‘Xinshang'Lily seed factory production.
