影响草地早熟禾原生质体生长和分裂的生理生化因素分析

摘  要:
以草地早熟禾‘午夜Ⅱ号’、‘新格莱德’和‘橄榄球Ⅱ号’3个品种继代20次的愈伤组织为材料,进行原生质体培养,测定愈伤组织可溶性蛋白质含量、氨基酸含量、SOD和POX酶活性.结果表明:3个草地早熟禾品种原生质体经培养15d后,原生质体分裂率分别为‘新格莱德’50.1%、‘午夜Ⅱ号’42.8%、‘橄榄球Ⅱ号’29.4%.前2个品种原生质体在培养过程中生长状态良好,持续分裂分化,而‘橄榄球Ⅱ号’原生质体在分裂数次后死亡.在愈伤组织更换新鲜培养基的第8~10d,3个品种可溶性蛋白质含量均达到各自动态变化曲线的最大值,分别为‘午夜Ⅱ号’2.672mg/g、‘新格莱德’2.834mg/g和‘橄榄球Ⅱ号’2.302mg/g.在培养的第8d,‘橄榄球Ⅱ号’愈伤组织中的氨基酸含量、SOD和POX活性均低于其他2个品种.

摘  要:
The callus subcultured for 20 times from the 'MidnightⅡ','Nuglade' and 'RugbyⅡ'which were varieties of Poa pratensis were used as material for protoplasts culture and its growth status were observed in this paper.Meanwhile,the soluble protein content and its content change with callus growth,amino acid content,SOD and POX activities were determined in the 3 varieties.The result indicated that after 15 d of protoplasts culture,the protoplasts from 'Nuglade' were found with the highest cell division rate(50.1%),followed by 'Midnight Ⅱ'(42.8%) and 'Rugby Ⅱ'(29.4%).'Nuglade' and 'Midnight Ⅱ' showed better growth behavior through whole process of protoplasts growth and sustained cell division and differentiation in these protoplasts happened,while protoplasts from 'Rugby Ⅱ' died after several times of cell division.After callus being cultured for 8 to 10 d by renewing new medium,the soluble protein contents in the 3 varieties all reached its maximum value.'Midnight','Nuglade' and 'Rugby Ⅱ' were at 2.672,2.834 and 2.302mg/g respectively.The amino acid contents,SOD and POX activities in callus from 'Rugby Ⅱ'were found lower than that of the other 2 varieties at the 8 th day of callus culture.
