5-aza-dc对延边奶山羊耳成纤维细胞生物学特性的影响

摘 要:
通过DNA甲基转移酶抑制剂5-aza-dc对延边奶山羊耳成纤维细胞进行不同处理后,利用MTT比色法选定5-aza-dc的最适作用浓度和时间,用Hochest33342/PI双染和琼脂糖凝胶电泳检测细胞凋亡情况,通过核型分析检测5-aza-dc对细胞染色体的影响。结果表明,低浓度的5-aza-dc(≤0.010μmol/L)作用72 h对细胞影响不大;随其浓度的增加,细胞形态、活率、凋亡及染色体都受到显著影响。5-aza-dc对细胞的最佳处理时间为72 h,此时低浓度的5-aza-dc并未引起细胞凋亡和核型改变。

摘 要:
To research the effects of DNA methylation inhibitor 5-aza-dc on biological characteristics of Yanbian dairy goat ear fibroblast cell,MTT method was used to determine the optimal action concentration and time of 5-aza-dc on Yanbian dairy goat ear fibroblast cells.PI and Hochest33342 double staining,agarose gel electrophoresis were used to detect apoptosis of the cells;and karyotype analysis to measure the effect of 5-aza-dc on the chromosomes.The results showed low concentration(≤0.010 μmol/L) of 5-aza-dC treatment for 72 h had little effect on the cells.With the increasing of concentration,cell morphology,viability,apoptosis and chromosomes were all significantly affected.The best processing time of 5-aza-dc was 72 h,low concentration of 5-aza-dc did not lead to apoptosis and karyotype change.
