华南地区猪圆环病毒3型感染的分子流行病学调查与分析

摘  要:
为了解猪圆环病毒3型(porcine circovirus 3,PCV3)在我国华南地区的感染情况,从2016年12月到2017年12月期间,从广东、广西、海南、福建、江西和湖南的177家规模化猪场采集1 078份病料,利用PCR技术检测PCV3的流行情况,并对PCV3阳性病料进行全基因组测序分析。结果显示:样品阳性率为29.04%(313/1 078),猪场阳性率为44.63%(79/177);从病料中测得的10株PCV3毒株全基因组核苷酸序列相似性为98.8%~100%,与国内外其他毒株的同源性在98.2%~100%之间;全基因组遗传进化分析显示PCV3遗传进化可以分为PCV3a和PCV3b,其中毒株CHN/HUNAN、CHN/GD-SH和CHN/GD-ZQ属于以美国毒株US/MO2016为代表的PCV3a分支,其余7个毒株属于PCV3b分支。结果表明:PCV3广泛存在于华南地区的猪群中,其基因组在不断进化,对PCV3的流行与致病机理有待进一步研究。

摘  要:
Porcine circovirus type 3( PCV3) is a novel emerging circovirus in recent years. The occurrence of PCV3 in South China remains unclear. In order to investigate the prevalence of PCV3 in South China,1 078 diseased samples were collected from 177 pig farms in Guangdong,Guangxi,Hainan,Fujian,Jiangxi and Hunan provinces through December 2016 to December 2017. PCR assay was conducted to detect the PCV3 genomic DNA. Positive samples were used to amplify and sequence the complete genome of PCV3. The PCR results demonstrated that the positive rate was 29. 04%( 313/1078) at the sample level and 44. 63%( 79/177) at the pig farm level. Complete genomic sequences were obtained from 10 PCV3 positive samples. The 10 complete genome sequences shared 98. 8% to 100% nucleotide identity between each other and 98. 2%-100% nucleotide identity when compared with other PCV3 strains available in Gen Bank. Phylogenetic analysis based on PCV3 complete genomes showed that 37 PCV3 strains divided into two clusters. PCV3 strains CHN/HUNAN,CHN/GD-SH and CHN/GD-ZQ belonged to PCV3 a clustering within US/MO2016,whereas the other 7 strains in this study belonged to PCV3b. This study indicated that PCV3 had been widely spreading in South China and further research on epidemiology and pathology should be conducted to comprehensively understand PCV3.
