肠出血性大肠杆菌强定殖株中新的双组份系统的鉴定

摘  要:
为了比较7株肠出血性大肠杆菌O157∶H7的定殖能力,采用灌胃感染的方式,将7株肠出血性大肠杆菌分别感染小鼠。对粪便中的细菌以及盲肠内定殖的细菌进行分离、计数,发现牛源菌株C1的定殖能力最强,且定殖维持时间最长。对C1菌株的基因组序列分析得到1对功能未知的双组份调节系统(TCS),N5512/N5520。PCR检测显示N5512/N5520存在于绝大多数肠出血性大肠杆菌O157∶H7临床分离株(98.5%),而在其他致病型的大肠杆菌中没有检测到。本研究为大肠杆菌O157∶H7的定殖和致病机制研究奠定了基础。

摘  要:
In order to compare the colonization capacity of seven strains of Enterohemorrhagic E. coli O157: H7,we challenged the mice by oral gavage of each individual strain. Bacteria were then isolated from the feces and cecum of the treated rodents,followed by proper counting of the bacteria. Our results showed that the cattle-derived strain C1 displayed the highest colonization,compared with the other strains. Sequence analysis of the C1's genome led to the discovery of a novel two-component signaling system,whose function remains unknown. Gene prevalence data demonstrated that N5512/N5520 existed in most of the Enterohemorrhagic E. coli O157: H7 clinical isolates( 98. 5%),but not in the other pathotypes of E. coli tested. Our findings here provide a preferred strain and a potential virulence-associated factor for future study on enterohemorrhagic Escherichia coli O157: H7 colonization and its virulence mechanisms.
