山羊重组IL-17A克隆表达与生物活性分析

摘 要:
为了分析山羊白介素17A(IL-17A)对山羊单核、中性粒细胞功能的影响,采用RT-PCR方法对山羊IL-17A基因进行克隆,并在体外表达得到重组蛋白质(rIL-17A),以不同质量浓度的重组蛋白rIL-17A(5、10、20、40μg/mL)与单核细胞和中性粒细胞共孵育,测定rIL-17A对山羊单核细胞和中性粒细胞的迁移、中性粒细胞凋亡的免疫调节效应。结果显示:5、20、40μg/mL的rIL-17A能够极显著促进山羊单核细胞的迁移(P<0.01),10μg/mL的rIL-17A也能显著促进山羊单核细胞的迁移(P<0.05);与中性粒细胞共孵育时,10μg/mL(P<0.05)和20、40μg/mL(P<0.001)的rIL-17A能显著或极显著促进山羊中性粒细胞的迁移;20μg/mL(P<0.05)、40μg/mL(P<0.001)的rIL-17A能显著或极显著诱导山羊中性粒细胞的凋亡。结果表明:重组蛋白rIL-17A具有良好的生物学活性,能在体外不同程度地促进山羊单核细胞、中性粒细胞的迁移及中性粒细胞的凋亡。

摘 要:
To analyze the effect of goat interleukin 17A on the function of goat monocytes and neutrophils,the IL-17A gene of goat was cloned by RT-PCR and the recombinant protein( rIL-17A) was expressed in vitro. Here,the immuno-modulatory effects of rIL-17A protein on monocyte and neutrophil migration and neutrophil apoptosis were investigated by co-incubation of rIL-17A at different concentrations( 5,10,20,40 μg/mL) with monocytes and neutrophils. The results showed that rIL-17A at the concentrations of 5,20,40 μg/mL significantly enhanced the migration of goat monocytes( P<0. 01),and rIL-17A at the concentration of 10 μg/mL also significantly promoted migration of goat monocytes( P<0. 05). In co-incubation with neutrophils,rIL-17A at the concentrations of 10( P<0. 05),20 and 40 μg/mL( P<0. 001) significantly promoted migration of goat neutrophils. rIL-17A at the concentrations of 20 μg/mL( P<0. 05) and 40 μg/mL( P<0. 001) significantly induced apoptosis of goat neutrophils. All these results suggested that the recombinant IL-17A protein had good biological activity and contributed to migration of goat monocytes,neutrophils and to apoptosis of goat neutrophils to varying degrees in vitro.
