NMHC-ⅡA在PRRSV感染Marc-145细胞过程中的作用

摘  要:
借用PRRSV结构蛋白GP5的抗独特型抗体Mab2-5G2,提取并纯化非肌肉肌球蛋白重链IIA(NMHC-ⅡA);通过激光共聚焦技术定位NMHC-ⅡA在PRRSV感染Marc-145细胞过程中的分布;并采用免疫共沉淀技术研究其与PRRSV受体CD163的相互作用。结果表明:NMHC-ⅡA在Marc-145细胞内的表达量随着细胞被病毒感染时间的延长而增加,当PRRSV完全进入细胞后,NMHC-ⅡA的表达量有所降低;NMHC-ⅡA与PRRS病毒受体CD163存在相互作用,作用位点在M1(20—259 aa)和M3(714—1 040 aa)上。试验证明,NMHC-ⅡA在PRRSV感染过程中可能起协同作用。

摘  要:
The non-muscle myosin heavy chain ⅡA(NMHC-ⅡA)was extracted and purified using the anti-idiotypic antibody Mab2-5 G2 of PRRSV structural protein GP5. The distribution of NMHC-ⅡA was located in theprocess of PRRSV infected Marc-145 cells by confocal laser technology. The interaction of NMHC-ⅡA withCD163 was studied by co-immunoprecipitation technology. The results showed that the expression of NMHC-ⅡAin Marc-145 cells increased with the prolongation of the time of virus infection. When PRRSV completely enteredthe cell,the expression of NMHC-ⅡA decreased. NMHC-ⅡA interacted with PRRS virus receptor CD163,theaction sites were located on M1(20—259 aa)and M3(714—1 040 aa). It was proved that NMHC-ⅡA mightplay a synergistic role in the process of PRRSV infection.
