大豆紫色酸性磷酸酶GmPAP17克隆与功能分析

摘  要:
土壤磷多以植物不能直接吸收利用的植酸盐形式存在,而紫色酸性磷酸酶具有分解利用植酸磷,提高磷素利用效率的功能。为进一步发掘该类基因,本研究利用磷高效大豆‘中黄15’与磷低效种质‘牛毛黄’,克隆并分析紫色酸性磷酸酶基因GmPAP17的生物学功能。结果发现:GmPAP17具有996bp开放阅读框,编码331个氨基酸残基,编码蛋白具有较高的酸性磷酸酶与植酸酶活性;植酸磷处理条件下,GmPAP17在磷高效大豆中的表达量显著高于低效种质,且以胁迫处理后30~42d差异最大,说明该基因与大豆开花结荚期磷素高效利用相关;进一步分析超表达转GmPAP17拟南芥发现,3个转基因株系在植酸磷胁迫下的植株生物重与磷浓度显著高于野生型对照,证实GmPAP17具有提高转基因拟南芥植酸磷利用效率的功能。

摘  要:
The majority of phosphorus exists in phytate form in soil,which could not be absorbed and used by plants directly.Purple acid phosphatase could hydrolyze the phytate phosphorus,which was significantly important for plants to improve the utilization efficiency of soil phosphorus.In view of this,one purple acid phosphatase gene named GmPAP17 was cloned and analyzed from the highand low-efficiency soybean varieties‘Zhong-huang 15'(ZH15)and‘Niu-mao-huang'(NMH)in this study.The results showed that GmPAP17 had a 996 bp ORF,and could encode 331 amino acids.The coding protein of GmPAP17 had a relative higher acid phosphatase and phytase activity.The transcriptional expression levels of GmPAP17 was significantly induced and much higher in ZH15 than NMH under the phytate treatment conditions,especially at 30—42 dafter treatments,which indicated that GmPAP17 involved in the phytate utilization during the period of flowering to podding in soybean.Furthermore,the plant weights and phosphorus concentrations of three over-expression transgenic Arabidopsislines were much significantly higher than the wild-type control,which demonstrated that GmPAP17 had the functions of improving the utilization efficiency of phytate phosphorus.
