野油菜黄单胞菌效应蛋白XopXccN抑制鞭毛蛋白诱导的MAPK激酶活性

摘  要:
野油菜黄单胞菌(Xanthomonas campestris pv.campestris,Xcc)通过III型分泌系统分泌效应蛋白到植物体内,帮助其在植物体内侵染和繁殖。XopXccN是野油菜黄单胞菌Xcc8004菌株的一个效应蛋白,利用同源重组双交换法将基因xop Xcc N在Xcc8004的基因组上做了删除突变,并对突变菌株进行了遗传互补。通过在拟南芥叶片上接种野生型Xcc8004、突变体菌株Δxop Xcc N以及互补菌株Δxop Xcc NC,发现效应蛋白XopXccN对于Xcc8004在拟南芥叶肉组织内的完整毒性是必须的。进一步研究发现,XopXccN可以抑制鞭毛蛋白诱导的MAPK激酶活性,其抑制发生在MKK下游。另外,XopXccN还可以抑制鞭毛蛋白诱导的下游标志基因FRK1(g22-induced receptor-like kinase)的表达。将XopXccN蛋白分段表达后发现,对于FRK1基因表达的抑制依赖于XopXccN的全长蛋白序列。本研究结果表明,XopXccN通过抑制M APK激酶活性,从而抑制寄主的先天免疫反应。

摘  要:
Xanthomonas campestris pv campestris( Xcc) is a causal agent of black rot diseases on numerous crucifer plants including Brassica and Arabidopsis. Xcc secretes a large repertory of effector proteins into host cells via type III secretion system to modulate host responses,enabling successful infection and multiplication in host tissue. Xop Xcc N is an effector protein of Xcc strain 8004,and belongs to the Xop N subfamily.Previous studies have show n Xop Xcc N mediates virulence function in host Chinese radish,and Xop Xcc N can interfere w ith photosystems, reactive oxygen species generation and callose deposition during Xcc8004 infection. How ever,it remains unknow n w hether Xop Xcc N suppresses pathogen-associated molecular patterns induced innate immunity. In this study,upstream and dow nstream flanking regions,320 bp from the xop Xcc N w ere PCR amplified and the tw o PCR fragments ligated into p K18 mobsac B. An xop Xcc N deletion strain( Xcc8004 Δxop Xcc N) was generated by two exchange steps using the plasmid p K18 mobsac B,and a complementation strain( Xcc8004 Δxop Xcc NC) w as created using a broad-hostrange vector p HM1 w ith the full xop Xcc N ORF. Well-know n,Arabidopsis-Xcc interaction has been used as a model pathosystem. Here,w hen infiltrated w ith WT Xcc8004,Xcc8004 Δxop Xcc N and Xcc8004 Δxop Xcc NC stains in Arabidopsis Col-0leaves,it w as found that Xop Xcc N w as required for Xcc8004 full virulence in mesophyll of Arabidopsis.Furthermore,Transient expression of Xop Xcc N in Arabidopsis protoplasts show ed that Xop Xcc N could suppress flg22-induced M APKs activity,and inhibited M APK activation triggered by the constitutive active M KK5 DD,suggesting that Xop Xcc N acts at MAPK cascades to inhibit PTI signaling. In addition,Xop Xcc N could also suppress flg22-induced FRK1∷LUC gene expression in Arabidopsis protoplasts. Truncated proteins assay show ed that the suppression of FRK1∷LUC transcription w as dependent on full length of Xop Xcc N. All the results suggest that Xop Xcc N can suppress the PTI signaling pathw ay,and the suppression may be in dow nstream of M KK5.
