白花草木樨种质苗期耐盐指标筛选及耐盐性综合评价

摘  要:
采用水培法,对48份白花草木樨幼苗进行NaCl(0(对照)、50、100、150、200 mmol/L)处理,测定了苗高(SH)、存活率(SR)、单株干质量(DW)、单株绿叶数(NGL)、生长速率(GR)和叶片叶绿素含量(Chl)6项指标,以各单项指标耐盐系数为基础,运用主成分分析、隶属函数法等方法进行综合评价;并采用逐步回归分析法建立了白花草木樨耐盐性预测回归方程。结果表明:NaCl处理后供试材料SH、SR、DW、NGL、GR和Chl 6项指标耐盐系数均低于对照,且各指标耐盐系数间存在显著相关性。通过主成分分析,本试验将NaCl处理下测定的6项指标转换成4个独立的综合指标,可代表供试材料92.02%的信息,结合隶属函数法分析,得到了48份种质材料耐盐性的综合评价值(D值);通过聚类分析,可将供试材料分为强耐盐型、中等耐盐型、弱耐盐型和敏盐型4类;进一步通过逐步回归分析,建立了回归方程D=-0.614+0.905X_(GR)+0.682X_(Chl)+0.435X_(NGL)(R~2=0.974),表明GR、Chl、NGL这3个指标对供试材料耐盐性有显著影响,在同等实验条件下,可通过测定GR、Chl和NGL这3项指标值对白花草木樨耐盐性进行预测,为白花草木樨种质资源耐盐性评价及新品种选育提供依据。

摘  要:
Forty eight Melilotus alba seedlings were exposed to combinations of NaCl(0, 50, 100, 150 and 200 mmol/L) in a hydroponics experiment, in which changes of seedling height(SH), survival rate(SR), dry weight(DW), number of green leaves(NGL), growth rates(GR) and chlorophyll content(Chl) were monitored. Based on salt tolerance coefficient of each single index, the salt comprehensive evaluation of 48 Melilotus alba accessions were evaluated by principal components analysis, subordination function method and cluster analysis. A regression equation of Melilotus alba salt tolerance was also established by stepwise regression analysis. The results showed that six indexes related to salt tolerance in experimental group were lower than those in control group. There were significant correlations among different materials. Based on multiple calculation methods(principal components analysis, subordination function method, cluster analysis and regression analysis), six indexes could be classified into four independent comprehensive components which may represent 92.02% of the raw data information. Integrated with subordinate function values analysis, the salt-tolerance comprehensive evaluation value(D value) of different accessions was obtained. The forty-eight accessions were divided into four salt-tolerant types(higher salt-resistant type, medium salt-resistant type, weaker salt-resistant type and salt-sensitive type) according to cluster analysis. The prediction equation for Melilotus alba salt tolerance(D=-0.614+0.905 X_(GR)+0.682 X_(Chl)+0.435 X_(NGL)) was established by stepwise regression analysis, which indicated that three indicators of GR, Chl and NGL had significant effect on the salt tolerance of tested materials. Therefore, the salt tolerance of Melilotus alba can be preliminarily evaluated by these three indexes(GR, Chl and NGL) under the same experimental conditions, which could provide important theoretical basis for evaluating germplasm resources and breeding new varieties of Melilotus alba.
