KT对白花草木樨不同外植体愈伤组织培养的影响

摘  要:
以改良HS为基本培养基,研究了白花草木樨不同外植体和KT(激动素)浓度对愈伤组织诱导的影响。结果表明:以无菌苗子叶、下胚轴和幼根为外植体,下胚轴出愈率为100.0%,为诱导愈伤组织的最佳外植体;子叶和下胚轴出愈率随KT浓度增大呈抑制作用,在1.0 mg/L 2,4-D+0.1 mg/L KT时出愈率分别为98.00%和100.00%,为最佳激素浓度配比;幼根出愈率随KT浓度增大先促进后抑制,1.0mg/L 2,4-D+0.3 mg/L KT时出愈率为99.33%,为最佳激素浓度配比。

摘  要:
The modified HS was used as a basal medium to study the effects of different explants of Melilotus suaveolens,including aseptic seedling of cotyledon,hypocotyl and radicle,and KT(kinetin) concentrations on callus induction.The results showed that the hypocotyl was the best explant for inducing callus and the callus rate of was 100.0%.The callus induction rates of cotyledon and hypocotyl were inhibited along with the increase ofKT concentration,and under the condition of 1.0mg/L 2,4-D+0.1mg/L KT,their callus rateswere 98.00% and 100.00%,respectively.As for radicle,its callus induction was increased along with the increase of KT concentration and then reduced.And under the condition of 1.0mg/L 2,4-D+0.3 mg/L KT,the callus rate was 99.33%.
