解淀粉芽胞杆菌固态发酵黄芪培养基的研究

摘  要:
为筛选优化解淀粉芽胞杆菌固态发酵黄芪培养基,用于生产发酵黄芪制品,以黄芪黄酮质量分数为指标,采用单因素试验对菌株适宜发酵培养基成分进行筛选;采用二次正交旋转组合试验设计及响应面法,优化黄芪固态发酵培养基。结果显示,在筛选成分中最适发酵培养基成分为黄芪、黄豆粉和碳酸钙。用其分别替代基础培养基中的相应营养物质,然后接种解淀粉芽胞杆菌,置37℃以150 r/min振荡培养24 h,其固态发酵物中黄芪黄酮质量分数分别为2.84、2.78和2.75 mg/g。优化的固态发酵培养基为:黄芪粉300 g/L、黄豆粉100 g/L、碳酸钙2 g/L、水598 m L/L,37℃发酵培养24 h,固态发酵物中的黄芪黄酮质量分数达3.21mg/g。结果表明,优化的黄芪固态发酵培养基适宜解淀粉芽胞杆菌生长,易于黄芪黄酮释放,筛选培养基原材料符合固态发酵产业化生产条件。

摘  要:
The aim of this study was to screen medium for solid fermenting Radix Astragalus by Bacillus amyloliquefaciens in order to producing fermented astragalus product.The effective components of solid fermentation medium were selected by the single factor experiments utilising the content of astragalus flavonoids as an indicator.The components of solid fermentation medium were optimised by quadratic orthogonal rotation combination design and response surface methodology.In results,the optimum components of medium were astragalus,soybean flour and calcium carbonate.The corresponding nutrients in basal medium were replaced by optimum components,then inoculated culture bacteria under the fermentation conditions at 37 ℃,150 r/min for 24 h,the content of astragalus flavonoids were 2.84 mg/g,2.78 mg/g and 2.75 mg/g,respectively.The optimum medium was consisted of 300 g/L astragalus,100 g/L soybean flour,2 g/L calcium carbonate and 598 m L/L water.The content of astragalus flavonoids of B.amyloliquefaciens cultured in the optimised medium for 24 h at 37℃ was 3.21 mg/g.In conclusion, the optimised solid fermentation medium was suitable for the growth of B.amyloliquefaciens,astragalus flavone release easily,materials are ideal for industrial production conditions of solid fermentation.
