偏凸山羊草fast ω-醇溶蛋白基因克隆及与WDEIA相关的IgE结合抗原表位分析

摘  要:
克隆偏凸山羊草中的fastω-醇溶蛋白基因,并对其序列和IgE结合抗原表位进行分析,以期了解IgE结合抗原表位在D~v和M~v基因组中的分布,并为制备单克隆抗体进行IgE结合抗原表位的定量分析奠定基础。结果表明,从偏凸山羊草中克隆获得8个fastω-醇溶蛋白基因(KY368672—KY368679),其中,3个fastω-醇溶蛋白(KY368672—KY368674)由D~v基因组编码,其余5个(KY368675—KY368679)由M~v基因组编码。序列分析发现,获得的fastω-醇溶蛋白与已发表的fastω-醇溶蛋白具有相似的序列结构,KY368672为SRQ型ω-醇溶蛋白,KY368673和KY368674为TRQ型ω-醇溶蛋白,KY368675—KY368679均为SRL型ω-醇溶蛋白。另外,在M~v基因组编码的fastω-醇溶蛋白中,发现了与α-醇溶蛋白相似的多聚谷氨酰胺序列。IgE结合抗原表位分析表明,M~v基因组编码的fastω-醇溶蛋白比D~v基因组编码的fastω-醇溶蛋含有更多的IgE结合抗原表位。

摘  要:
The fast ω-gliadin gene of Aegilops ventricosa was cloned,and its sequence and IgE-binding epitopes were analyzed,so as to understand the distribution of IgE-binding epitopes in D~v and M~v genomes and lay the foundation for the quantification of IgE-binding epitopes by preparation of monoclonal antibody.The results indicated that eight novel fast ω-gliadin genes were cloned from Aegilops ventricosa,and the number of fast ω-gliadins were KY368672—KY368679.Three fast ω-gliadins(KY368672—KY368674) were encoded by the D~v genome,and the other five fast ω-gliadins(KY368675—KY368679) were encoded by the M~v genome.Sequence analysis showed that the sequence structures of all fast ω-gliadins were similar to other published fast ω-gliadins.KY368672 was SRQ type ω-gliadin,KY368673 and KY368674 were TRQ type ω-gliadins,and KY368675—KY368679 were SRL type ω-gliadins.The fast ω-gliadin encoded by the M~v genome had similar polyglutamine sequence to that in α-gliadin.IgE-binding epitope analysis indicated that the fast ω-gliadin encoded by the M~v genome contained more IgE-binding epitopes than those encoded by the D~v genome.
