烟草花叶病毒侵染对光合组织中蛋白质的影响及其防治研究

摘  要:
为了提高烟草对烟草花叶病毒(TMV)侵染的耐受性,筛选有效防治TMV的制剂,采用i TRAQ技术探寻TMV侵染烟草后不同时间段蛋白质组的变化。结果显示:光合系统、卟啉代谢和叶绿素代谢途径中PSⅡ32 ku protein、PSⅠP700 apoprotein A1、Cytochrome b/f complex subunitⅣ、Oxygen-evolving enhancer protein 1、Geranylgeranyl diphosphate reductase及Short-chain dehydrogenase TIC 32的表达量在TMV侵染后发生显著变化。在此基础上,筛选了对植物叶片组织具有保护作用的复配制剂A和B。采用qRT-PCR检测结果显示,制剂B提高了烟株光合途径和卟啉代谢与叶绿素代谢途径中关键基因(Oxygen-evolving enhancer protein 1、Short-chain dehydrogenase TIC32)的表达水平,降低了TMV对光合系统的破坏。进一步检测抗性基因N和PR1-a的表达量,并结合半叶枯斑试验,均表明制剂B对烟草花叶病具有显著的防治效果。

摘  要:
In order to improve the tolerance of tobacco to tobacco mosaic virus( TMV) infection,and screen effective preparations to control TMV,i TRAQ technology was used to explore proteome changes in different time after TMV infection of tobacco. It was found that the protein expression level of PSⅡ 32 ku protein,PSⅠP700 apoprotein A1,Cytochrome b/f complex subunit Ⅳ,Oxygen-evolving enhancer protein1,Geranylgeranyl diphosphate reductase and Short-chain dehydrogenase TIC 32 in photosynthetic system,porphyrin metabolism and chlorophyll metabolism pathway significantly changed with the prolongation of TMV infection time. Anti-TMV compound preparations A and B were selected to treat tobacco leaves,and the expression levels of these genes as well as resistance genes were detected by qRT-PCR. The results showed that the anti-TMV preparation B significantly promoted the expression of Oxygen-evolving enhancer protein 1 and Short-chain dehydrogenase TIC 32 and reduced the damage of TMV to photosynthetic system. Further detection of lesions by half-leaf method and expression levels of N and PR1-a genesshowed that TMV infection was significantly inhibited by the preparation B.
