不同种源印楝遗传多样性的ISSR分析

摘  要:
利用ISSR分子标记技术对印楝进行遗传多样性分析,用9条引物进行扩增,共检测出81条清晰的位点,其中79条具有多态性,多态位点百分率为97.53%,Ne i s基因多样性指数为0.380 3,Shannon信息指数为0.553 7,表明印楝遗传多样性很丰富。种源间遗传分化系数为0.470 2,Shannon s居群分化系数为0.467 7,表明印楝种源内的遗传分化大于种源间的分化。聚类分析将13个种源分为2大类,来自缅甸的11个种源聚成一类,来自澳大利亚和印度的种源聚成另外一类。结果表明,ISSR分子标记技术适合于印楝的遗传多样性分析。

摘  要:
The genetic diversity of neem was analyzed by ISSR technique.9 appropriate primers were selected for ISSR PCR amplification.The 9 primers could amplify 81 bands.Of these,79 were polymorphic loci.The results showed that high levels of genetic diversity existed in the populations(PPB was 97.53%,H was 0.380 3 and I was 0.553 7).Coefficient of gene differentiation Gst was 0.470 2,Shannon's diversity index was 0.467 7,which showed that the genetic variation of neem mainly came from intra-provenance.The cluster analysis showed that thirteen provenances were divided into two clusters.One was consisted of eleven provenances of Burma,and the other was Australia and India provenances.This fact indicated that eleven Burma provenances have a close genetic relationship,so do Australia and India provenances.The results showed that ISSR method was suitable for genetic diversity analysis of Neem,which provide the scientific basis of developing effective protection measures for these species.
