中国芦荟AlDREB2基因的克隆及其胁迫表达

摘  要:
为了研究中国芦荟的抗逆机理,以cDNA为模板,利用3′RACE和PCR扩增方法,克隆得到了一个编码DREB蛋白的基因,命名为AlDREB2,GenBank登录号为FJ560460。其cDNA序列全长为886bp,包含一个636bp的开放阅读框,推导编码的氨基酸序列(212个氨基酸)与库拉索芦荟AlDREB1的序列相似性很高。进化树分析结果将AlDREB2归入DREB亚家族中A-6亚组。利用RT-PCR技术分析了AlDREB2基因的表达与胁迫的关系,表明AlDREB2基因在非生物胁迫与生物胁迫中能够被不同程度诱导表达。

摘  要:
DREB (dehydration responsive element-binding factor) genes encode a small family of transcriptional activators functioning in plant tolerance and acclimation to various stresses. To investigate the molecular mechanisms underlying stress resistance in aloe (Aloe vera L. var. chinensis),a new gene encoding DREB protein was cloned from aloe via combined method of PCR and 3'RACE. Based on the high homology to previous reported AlDREB1 ,this gene (GenBank accession number,FJ560460) was named as AlDREB2. Sequence alignment analysis located AlDREB2 to A-6 sub-group of DREB family. RT-PCR revealed that expression of AlDREB2 was significantly induced by ABA and dehydration treatments (salt or PEG6000),though with different dynamic patterns. Besides,transcript accumulation of AlDREB2 was up-regulated by cold stress and external ethylene. These results best support the interpretation that AlDREB2 is an active regulator involved in dehydration tolerance and cold acclimation in aloe.
