摘  要:
本试验旨在建立基质分散固相萃取净化技术在荧光检测条件下测定饲料中香芹酚和百里香酚含量的高效液相色谱分析方法。饲料使用水和正己烷振荡提取,提取液中的香芹酚和百里香酚经复合吸附材料萃取后,使用乙腈洗脱。以Eclipse Plus C18色谱柱(250 mm×4.6 mm,5μm)为固定相,乙腈/水(65∶35,V/V)等度洗脱,流速1.0 mL/min,荧光检测器激发波长274 nm,发射波长304 nm,外标法定量。基于该法对不同饲料基质进行分析测定,香芹酚和百里香酚的含量在1~500 ng/mL线性关系良好(线性系数≥0. 999 0),方法检出限为0.6μg/kg,定量限为2.0μg/kg。在2.0、10.0和20.0μg/kg 3个添加水平下,香芹酚和百里香酚的回收率为80.2%~108.6%,其相对标准偏差(RSD)小于10.2%。该方法操作简单,灵敏度高,结果准确可靠,可用于饲料中香芹酚和百里香酚含量的分析测定。

摘  要:
A high performance liquid chromatography method was developed for the determination of carvacrol and thymol contents in feed by matrix solid-phase dispersion technology under fluorescence detector. With water and n-hexane used as extraction solvent,the carvacrol and thymol in the extraction solution were extracted by the multiple adsorption material and eluted by acetonitrile. Analysis was performed on an Eclipse plus C18 column(250 mm×4.6 mm,5 m)with the mobile phase consisting of acetonitrile/water(65∶35,V/V)at a flow rate of 1.0 mL/min. The excitation wavelength of the fluorescence detector was 274 nm,and the emission wavelength was 304 nm. Different feed samples were analyzed by the established methods. Good linearity was obtained in the ranges of 1 to 500 ng/mL,with linear coefficients(R2)higher than 0.999 0. The limit of detection(LOD)was 0.6μg/kg and the quantification limit(LOQ)was 2.0μg/kg. The recoveries for carvacrol and thymol spiked(2,10 and 20μg/kg)in feeds ranged from 80.2% to 108.6% and the relative standard deviations(RSD)were lower than 10.2%.
