烟叶样品中18种氨基酸的微波水解分析方法研究

摘  要:
氨基酸在烟叶生长过程的不同阶段会发生不同的变化,影响烟叶品质。为控制烟叶质量,快速、准确测定烟叶氨基酸,本文采用了微波水解烟叶样品,氨基酸自动分析仪钠离子系统测定18种氨基酸。通过微波水解的条件试验确定了微波水解烟叶最佳条件:烟叶称样量30~60 mg,水解温度160℃,保温时间20 min,该方法测定18种氨基酸回收率在70.1%~109.4%,与常规酸水解方法回收率相当,准确度高。相对标准偏差0.83%~10.10%与常规酸水解法接近,该方法快捷方便、节能、准确,适合烟叶水解氨基酸的测定。

摘  要:
Amino acid contents,which affect the quality of tobacco,vary during the different stages of tobacco growth process. In order to quantify the amino acids fast and accuratly for quality control of tobacco,herein,18 amino acids of tobacco samples were analyzed with sodium ion system of automatic amino acid analyzer and microwave hydrolysis. The optimum condition for microwave hydrolysis of tobacco amino acids was obtained from condition tests: sample weight 30- 60 mg,hydrolysis temperature 160 ℃,holding time 20 min. The average recoveries and relative standard deviations were 70. 1 %- 109. 4 %,and 0. 83 %- 10. 1 %,respectively,which were comparable with conventional acid hydrolysis method. The proposed method is facial,accurate,energy saving and suitable for the hydrolytic amino acids determination of tobacco.
