假俭草体细胞抗寒突变体的筛选及其鉴定

摘  要:
假俭草抗寒性差是限制其广泛应用的主要因素之一。本研究目标是以优良假俭草选系‘翠绿1号’为材料,经低温诱导和筛选获得体细胞抗寒突变体。实验结果表明:98块假俭草种子诱导的愈伤组织在继代培养的过程中,经0℃的低温条件下培养24 d,获得了1块存活的愈伤组织,该愈伤组织经过继代增殖后,进行分化、生根和移栽,获得大量的突变体植株。半致死温度的结果表明:突变体和对照叶片半致死温度(LT50)分别为-6.946℃和-5.851℃,处理与对照之间达到显著性差异。SRAP的结果表明:假俭草突变体在200 bp处存在特征带,表明假俭草体细胞突变体的变异性在分子水平上与对照存在差异。因此,本试验为植物抗寒突变体的筛选提供了有效的方法。

摘  要:
Poor cold-resistance of centipedegrass is one of the major factors limiting its wide application.The objective of this study is to obtain cold-resistant somatic mutants from the seed of'Cuilǜ No.1' centipedegrass by induction and screening at low temperature.The callus induced by seeds of centipedegrass was cultivated at the low temperature of 0℃for twenty-four days during the process of subculture and three pieces of survival callus tissues were obtained.somatic mutant is obtained after survival callus subculture proliferation,differentiation,rhizogenesisas well as transplantation.Somatic mutant and the control.LT50 of were-6.946℃and-5.851℃,respectively,which was significant difference.The results of SRAP markers demonstrated that there is characteristic bands at 200 bp of somatic mutant and indicated that the somatic mutants of centipedegrass were genetically stable comparing with that of the control plant on molecular level.This experimentation may provide effective means for screening of cold-resistance somatic mutant of plants.
