高山笃斯越桔离体快繁体系建立及种质试管保存研究

摘 要:
以高山笃斯越桔新生嫩芽为外植体,应用均匀设计法筛选其最适合的基部直接再生芽苗、生根及种质试管保存的培养基,结果表明,最适合的基部直接再生芽苗诱导培养基为:DR+TDZ2.0 mg.L-1+IBA0.01 mg.L-1,分化率为98.8%以上;生根培养基:MS(改良)+IAA0.5 mg.L-1+IBA0.07 mg.L-1+Kt0.1 mg.L-1,生根率达98%;试管保存培养基:1/5MS+B91.8 mg.L-1+根皮苷3.3 mg.L-1,保存时间可达43个月以上。以再生植株的茎节为材料进行快繁的结果表明,在35 d的一个培养周期内,增殖倍数平均达60以上。常温条件下,采取"低营养矮化延缓生长"的方法在试管内保存种质资源。建立了高山笃斯越桔的离体培养和种质试管保存体系。

摘 要:
The tender buds of Vaccinium uliginosum L.var.alpinum Nakai.were used as explants for the experiment.Uniform design for the most suitable media for shoot regeneration immediately at base of tender buds,rooting and germplasm preservation in vitro was screened.The results showed that DR+TDZ2.0 mg·L-1+IBA 0.01 mg·L-1 was fits for shoot regeneration,the rate of regeneration was more than 98.8%;MS(modified)+IAA0.5 mg·L-1+IBA0.07 mg·L-1+Kt0.1 mg·L-1for rooting,the rate of rooting was 98%;1/5MS+B91.8 mg·L-1+phloridzin3.3 mg·L-1for germplasm preservation in vitro for 43 months.Stems each with one node were cut from regenerated shoots and cultured for propagation,and a 60-fold proliferation rate was achieved within 35 days.The method of "low nutrients with defering growth and dwarfing" was utilized for germplasm peservation in vitro at normal temperature.In vitro culture and germplasm peservation in vitro system of Vaccinium uliginosum L.var.alpinum Nakai.has been successfully established.
