中药复方对骨髓抑制小鼠血液生理指标及细胞因子的影响

摘  要:
通过腹腔注射环磷酰胺建立骨髓抑制小鼠模型,采用血细胞自动分析仪测定模型小鼠及使用中药复方(剂量分别为20、10和5g/kg)、黄芪多糖(66mg/kg)3d和7d小鼠的血液生理指标,用ELISA方法测定血清中白介素-3、白介素-6和促红细胞生成素的含量。结果显示,按80mg/kg剂量腹腔注射环磷酰胺,每日1次,连用3d,可成功建立骨髓抑制小鼠模型;模型小鼠的白细胞总数、中性粒细胞总数、淋巴细胞总数、血小板总数及血清白介素-6、促红细胞生成素的含量显著降低;红细胞总数、血红蛋白含量及白介素-3的含量降低,但变化不显著;黄芪多糖及各剂量中药复方连续灌胃3d或7d,均可改善模型小鼠的血液生理指标,提高血清白介素-3和促红细胞生成素含量。连续用药3d,以中药复方20g/kg剂量的效果为最佳;连续用药7d,以中药复方10g/kg剂量的效果为最佳。按10g/kg剂量使用中药复方,连续灌胃7d,可极显著提高模型小鼠的白细胞总数、中性粒细胞总数、中性粒细胞百分比、淋巴细胞总数、中间细胞总数、中间细胞百分比、血小板总数以及血清白介素-3、促红细胞生成素的含量(P<0.01);抑制环磷酰胺所致的红细胞总数、红细胞压积、血红蛋白和白介素-6含量降低(P<0.01)。证实由鱼腥草、蒲公英、陈皮、苍术、白芍、白术和当归等组成的中药复方,具有改善环磷酰胺所致骨髓抑制小鼠造血功能的作用,效果优于黄芪多糖;中药复方按20g/kg剂量使用具有作用快速的特点,按10g/kg剂量使用作用稍缓但药效持久。

摘  要:
A model of myelosuppressive mice was established by intraperitoneal injection of cyclophosphamide.The blood physiological parameters of the model mice,and those treated with Chinese herbal compound(doses of 20 g/kg,10 g/kg and 5 g/kg) and Astragalus polysaccharide(66 mg/kg) for 3 days or 7 days,were measured by automatic blood cell analyzer.Meanwhile,the content of interleukin-3(IL-3),interleukin-6(IL-6) and erythropoietin was measured by ELISA.The results showed that the model of myelosuppression could be successfully established by daily intraperitoneal injection of 80 mg/kg cyclophosphamide for 3 days.The white blood cell count,neutrophile granulocyte count,lymphocyte count,platelet count and the content of IL-6 and erythropoietin decreased significantly in the myelosuppressive mice.Red blood-cell count,and content of hemoglobin and IL-3 decreased slightly. The blood physiological parameters and the content of IL-3 and erythropoietin could be improved to different degrees in the myelosuppressive mice by intragastric administration of Astragalus polysaccharide and each dose of the herbal compound for 3 days or 7days;the effect of 20g/kg dose of herbal compound was the best after medicated for 3 days;while the 10g/kg dose was the best for 7 days. After the myelosuppressive mice receiving daily intragastric administration of 10g/kg herbal compound for 7 days,the white blood cell count,neutrophile granulocyte count,neutrophile granulocyte percentage,lymphocyte count,intermediate cell count,intermediate cell percentage,platelet count,and the content of IL-3 and erythropoietin could be improved significantly(P<0.01),and the reduction of red blood cell count,hematocrit,and content of hemoglobin and IL-6 induced by cyclophosphamide could be inhibited significantly(P<0.01). The results indicated that the Chinese herbal compound composed by Houttuynia,Taraxacum mongolicum,dried tangerine peel,Atractylodes,white peony root,Atractylodes macrocephala,Angelica and other 7 Chinese medical herbs could improve hematopoietic function in myelosuppressive mice,and it was better than Astragalus polysaccharide;a 20g/kg dose had a role to take an effect fast and the 10g/kg dose slow but sustained longer.
