摘 要:
用农抗5102产生菌的两株不同营养缺陷型突变体WNF-2-1-90和WNF-2-1一135所进行的原生质体融合条件试验。结果表明,1000、4000和6000三种分子量的PEG,各以30%、40%和50%的浓度加入两株突变体的混合原生质体悬液,其融合率达1.8-4.1%,比不加PEG的提高2-7倍;较佳的PEG浓度,则因其分子量不同而异,1000PEG的诱导效果随加入浓度升高而增大,余二者则相反。混合原生质体悬液在4℃和28℃中放置4小时后,其融合率几乎和加PEG的相近。融合前立即用UV照射混合原生质体悬液,其融合率比未照射的提高2倍以上。比较直接选择法和间接选择法检出融合子,表明间接法比直接法检出的要高1倍以上。

摘 要:
This paper describes some conditions of protoplast fusion between twoauxotro phically markcd parental strians, WNF-2-I-90 and WNF-2-1-135, of antibioticproducing strain 5102. The results showed: I. Solutions of PEG of different molecularweight (e. g. 1000 up to at least 6000) were distinctly effective on indued protoplastfusion. A suitable concentration of PEG relate to its molecular weight. 2. Afterstorage of mixed protoplast suspensions of two parental strains at 4°C or 28°Cfor 4 hr. in abserce of PEG, the fusion frequency was almost similar to that of inprescence of PEG. 3. The fusion frequency of mixed protoplast suspensions treated bythe ultraviolet irradiation was 2 times higher than without ultraviolet irradiation. 4.The fusion frequency obtained by indirect selection method was 2×than that of directselection method.
