中国李Nubiana叶柄离体培养再生植株

摘  要:
以中国李Nubiana试管苗幼嫩叶片上的叶柄为试材,研究了基本培养基、激素种类配比、暗培养时间、培养基中琼脂浓度等对叶柄再生的影响,建立了其叶柄再生体系。结果表明,中国李Nubiana叶柄再生不定芽最佳培养基为WPM+TDZ 0.5 mg.L-1+蔗糖30 g.L-1+琼脂6 g.L-1,不定芽再生率可达到35.0%;暗培养为叶柄不定芽再生的必要条件,暗培养14 d后转入光下培养,能促进叶柄产生不定芽。中国李Nubiana叶柄再生表现出明显的极性,不定芽一般出现在叶柄的近轴端。

摘  要:
Experiment was conducted with the young petiole in vitro of Nubiana(Prunus salicina) for studying the effects of different basic culture media,hormone levels,dark culture and agar content on the adventitious shoot regeneration.The re-sults showed that the suitable basic medium and plant growth regulator was WPM +TDZ 0.5 mg.L-1+sucrose 30 g.L-1+agar 6 g.L-1 with 35.0% of regeneration rate.The dark was necessary for the petioles to regeneration.Dark culture for 14 days prior to light culture was helpful to adventitious shoot regeneration.Adventitious shoots were mainly regenerated at proximal cutting region.
