激光共聚焦扫描显微镜数据采集与分析技术研究

摘  要:
以梨花粉与花柱为试材,探索出了一套方便实用的激光共聚焦扫描显微镜(Laser Confocal Scanning Micro- scope,LCSM)数据采集与分析方法。利用这套方法可使在LCSM系统上进行数据处理的时间缩减到最少,而大部分 的工作可以带出实验室到一般的计算机上进行处理,从而大大降低实验费用。详细介绍了在梨花粉与雌蕊信号转导 机制的研究上如何利用普通计算机进行LCSM图像堆栈的读取、增加/去除伪彩、"蒙太奇"处理、Time Course数据转 化与数据处理等的方法与技术,以便为利用LCSM开展果树细胞信号转导的研究提供参考。

摘  要:
In this article, the author provided a convenient and practical method for collecting and processing the data gained from Laser Confocal Scanning Microscope (LCSM) with pollen and pistil of pear, through which the time needed for processing data in LCSM will be largely decreased and the most work can be completed with common computer, so that the expenses will be mostly reduced. The capturing of image stack, adding and discarding of pseudo color, treating with montage, and transducing and analyzing time course data with common computer were introduced in details, aimed to provide information for researchers who worked with LCSM in the field of signal transduction in the fruit trees.
